以醋酸叔丁酯和N-CBZ-L-天冬氨酸為原料,合成(S)-3-(((芐氧基)羰基)氨基)-4-(叔丁氧基)-4-氧代丁酸、N-芐氧羰基-L-天門冬氨酸4-叔丁酯及Z-L-天冬氨酸叔丁酯叔丁酯的一般步驟如下:按照實施例1中所述的方法建立酯交換反應體系,但將BF3·Et2O替換為其他催化劑。測試的催化劑包括酸類或鹽類,具體如表I所列。反應在室溫或約50℃條件下進行,反應時間為5.5至15小時。反應結束后,采用HPLC技術對反應混合物進行分析,分析方法同實施例1。表I結果表明,在濃硫酸(H2SO4)、甲磺酸(MsOH)、氯化鋅(ZnCl2)及二氯甲烷中的四氯化鈦(TiCl4 in DCM)等催化劑存在下,酯交換反應能顯著生成Z-Asp(OtBu)2。相比之下,氯化鋁(AlCl3)、氯化鐵(FeCl3)及四異丙醇鈦(Ti[OiPr]4)即使在提高反應溫度和延長反應時間的條件下,催化效果仍不顯著。
