以2-氨基-3-羥基吡啶(90.1 mmol)和原甲酸三乙酯(720.7 mmol)為原料合成噁唑并[4,5-b]吡啶的一般步驟:在氮?dú)獗Wo(hù)下,將2-氨基-3-羥基吡啶溶解于原甲酸三乙酯中,加入對甲苯磺酸(4.50 mmol)作為催化劑。將反應(yīng)混合物在160℃下攪拌反應(yīng)過夜。反應(yīng)完成后,通過減壓蒸餾除去過量的原甲酸三乙酯。粗產(chǎn)物采用柱色譜法進(jìn)行純化,洗脫劑為環(huán)己烷/乙酸乙酯(5%-40%梯度洗脫),得到噁唑并[4,5-b]吡啶,為米色固體,收率為68%。產(chǎn)物經(jīng)質(zhì)譜(LRMS)檢測,[M + H]+ m/z為121.1;1H NMR(DMSO-d6, 400 MHz)δ 9.04(s, 1H), 8.58(dd, J = 4.8, 1.4 Hz, 1H), 8.26(dd, J = 8.2, 1.4 Hz, 1H), 7.50(dd, J = 8.2, 4.8 Hz, 1H), 6.41(s, 2H);13C NMR(DMSO-d6, 101 MHz)δ 157.7, 154.5, 147.1, 142.0, 121.5, 120.1。
