以 (R)-1-(叔丁氧羰基)哌啶-3-羧酸為原料合成 3-(羥甲基)哌啶-1-甲酸叔丁酯的一般步驟如下: 實(shí)施例58:N-(2-甲基丙基)-N-(3,5-二氯苯基甲基)-(3R)-哌啶-3-基甲胺1.5L-酒石酸鹽的合成: (i)在室溫下,將硼烷-四氫呋喃絡(luò)合物(1M的THF溶液,65.4ml,65.4mmol,3當(dāng)量)緩慢滴加到 (R)-N-Boc-哌啶-3-羧酸(5g,21.8mmol,1當(dāng)量)的THF(50ml)溶液中。反應(yīng)混合物在室溫下攪拌16小時(shí)。隨后,將溶液冷卻至0℃,并小心加入2N氫氧化鈉水溶液(250ml)進(jìn)行水解。水解后的混合物加熱回流48小時(shí)。反應(yīng)完成后,將混合物再次冷卻至0℃,并用乙醚(3×100ml)萃取。合并有機(jī)層,用鹽水(100ml)洗滌,硫酸鎂干燥,過濾并真空濃縮。殘余物通過快速色譜法純化,使用50-70%乙酸乙酯的異己烷溶液作為洗脫劑,得到標(biāo)題化合物 (3R)-3-(羥甲基)哌啶-1-羧酸叔丁酯(4.56g,收率98%)。 1H NMR(300MHz,CDCl3)δ:4.0-2.7(6H,br m),2.17-1.19(5H,br m),1.46(9H,s)。
