通用方法:在攪拌條件下,將1-((2R,3R,4S,5R)-3,4-二羥基-5-(羥甲基)四氫呋喃-2-基)嘧啶-2,4(1H,3H)-二酮(5, 0.103 g, 0.4 mmol)溶解于乙腈水溶液(H2O:CH3CN = 1:9, 5 mL)中。隨后,加入NaN3(0.104 g, 1.6 mmol),并在室溫下加入SMBI(0.101 g, 0.44 mmol)。持續(xù)攪拌反應(yīng)混合物,并通過薄層色譜(TLC)監(jiān)測反應(yīng)進度。反應(yīng)1.5小時后,將反應(yīng)混合物過濾,減壓濃縮,并與乙腈(2×2 mL)共蒸發(fā)以去除殘余水分。粗產(chǎn)物通過柱色譜法純化(洗脫劑:二氯甲烷中的4%-6%甲醇,v/v),得到純品5-溴尿苷(6, 0.117 g, 93%),為白色固體。
