以2-(甲氧基羰基)-3-甲基吡啶-1-烯-1-醇鹽(500 mg,2.99 mmol,1.00當(dāng)量)為原料,將其溶解于磷酰氯(2 mL)中。將反應(yīng)混合物在110℃下攪拌反應(yīng)4小時(shí)。反應(yīng)完成后,用冰水淬滅反應(yīng),調(diào)節(jié)pH至7。隨后,用乙酸乙酯(3 × 10 mL)萃取水相。合并有機(jī)層,用無(wú)水硫酸鈉干燥,減壓濃縮除去溶劑。通過(guò)硅膠柱色譜法純化粗產(chǎn)物,以乙酸乙酯/石油醚(1:5,v/v)為洗脫劑,得到6-氯-3-甲基吡啶-2-羧酸甲酯(230 mg,收率41%)為白色固體。LC-MS(ESI,m/z):186 [M + H]+。
