以D-天門冬氨酸和氯甲酸芐酯為原料合成Cbz-D-天冬氨酸的一般步驟如下: 1. 在0℃下,向500 mL蒸餾水中加入6 M NaOH溶液,調(diào)節(jié)溶液的pH至13。隨后,加入65 g (489 mmol)的(R)-天冬氨酸。 2. 在磁力攪拌下,向上述溶液中緩慢加入141 g (831 mmol, 1.7當(dāng)量)的氯甲酸芐酯。 3. 將反應(yīng)混合物在室溫下攪拌反應(yīng)2天。 4. 反應(yīng)完成后,用乙醚洗滌混合物,水相用6 N HCl酸化至pH<2。 5. 酸化后的水相用乙酸乙酯(AcOEt)萃取,合并有機(jī)相。 6. 有機(jī)相用無(wú)水Na2SO4干燥,過(guò)濾后,在旋轉(zhuǎn)蒸發(fā)器中濃縮,得到80 g透明無(wú)色膠狀產(chǎn)物,產(chǎn)率為59%。 產(chǎn)物表征: - MS (ESI) m/e (MH+): 266 - 1H-NMR (CDCl3, 400 MHz): δ 7.21-7.21 (m, 5H), 6.14 (d, J = 8.4 Hz, 1H), 5.05 (s, 2H), 4.60 (m, 1H), 2.99 (m, 1H), 2.75 (m, 1H)。
