實(shí)施例4:化合物4(7-硝基-1H-吲哚)的制備;將化合物3(7-硝基吲哚-2-羧酸,1.63 g,7.91 mmol)溶解于喹啉(13 mL)中,隨后加入氧化銅(CuO,0.19 g)。將反應(yīng)混合物攪拌并加熱至194℃。在加熱過程中,觀察到氣體逸出。反應(yīng)2小時(shí)后,氣體停止產(chǎn)生,通過薄層色譜(TLC)確認(rèn)反應(yīng)完成。將反應(yīng)液緩慢倒入由濃鹽酸(21.3 mL)和冷水(42.6 mL)配制的稀鹽酸溶液中,攪拌后生成黑色沉淀。過濾反應(yīng)混合物,分離黑色沉淀,濾液用乙醚萃取。合并有機(jī)相,依次用飽和碳酸氫鈉(NaHCO3)溶液和去離子水洗滌。有機(jī)層經(jīng)無水硫酸鈉(Na2SO4)干燥后,過濾并濃縮,得到黃色固體。將該固體在乙醇(EtOH)中重結(jié)晶,得到針狀淺黃色固體,即化合物4(7-硝基-1H-吲哚,0.87 g,收率69%)。熔點(diǎn)96-97℃(文獻(xiàn)值95-96℃);1H NMR(200 MHz, CDCl3)δ 6.63(dd, J = 2.2, 1.0 Hz, 1H, ArH),7.08-7.18(m, 1H, ArH),7.32(t, J = 3.0 Hz, 1H, ArH),7.90(d, J = 7.8 Hz, 1H, ArH),8.08(d, J = 8.1 Hz, 1H, ArH),9.88(s, 1H, NH);13C NMR(50 MHz, 丙酮-d6)δ 103.7, 118.9, 119.2, 128.4, 129.1, 129.3, 132.9, 133.5;質(zhì)譜(EI)m/z 162(M+, 100%),116(M-46, 87%),104(M-58, 41%),89(M-73, 53%)。
