以6-硝基-異吲哚啉-1-酮為原料合成6-氨基-異吲哚啉-1-酮的一般步驟:將中間體6B(1.6 g,8.98 mmol)和Pd/C(0.18 g)在甲醇(100 mL)中的懸浮液在氫氣(1 atm)氛圍下攪拌4小時(shí)。反應(yīng)完成后,過(guò)濾反應(yīng)混合物,并用甲醇洗滌濾餅。合并濾液后,通過(guò)減壓濃縮除去溶劑。粗產(chǎn)物用甲醇(10 mL)研磨,隨后減壓干燥,得到中間體6(800 mg,5.40 mmol,產(chǎn)率60.1%),為米色固體。產(chǎn)物結(jié)構(gòu)經(jīng)1H NMR(400 MHz, DMSO-d6)確認(rèn):δ 4.15(s, 2H), 5.26(s, 2H), 6.77(dd, J = 8.25, 2.20 Hz, 1H), 6.80(s, 1H), 7.16(d, J = 8.79 Hz, 1H), 8.29(s, 1H)。質(zhì)譜(ESI)顯示m/z 149.2([M + H]+)。
