以5-溴-2-溴甲基苯甲酸甲酯為原料合成6-溴異吲哚啉-1-酮的一般步驟如下:在室溫下,向5-溴-2-溴甲基苯甲酸甲酯(1當(dāng)量)的1:1 THF/MeOH混合溶液中溫和鼓泡氨氣40分鐘。反應(yīng)完成后,將反應(yīng)混合物在真空下濃縮。將濃縮后的殘余物在CH2Cl2中超聲處理15分鐘,隨后過濾,得到目標(biāo)產(chǎn)物6-溴-2,3-二氫-異吲哚-1-酮,為白色固體(收率98%,純度90%)。產(chǎn)物質(zhì)譜數(shù)據(jù):m/z (LC-MS, ESP): 212.3/214.3 [M + H]+,保留時(shí)間R/T = 2.98分鐘。
