步驟2:將1-(4-溴-2-氟苯基)乙醇(5.9g,26.9mmol)溶解于110mL無水丙酮中,冷卻至0℃。在攪拌下,逐滴加入瓊斯試劑(由26.72g三氧化鉻溶于23mL濃硫酸中,再用水稀釋至總體積為100mL;使用6.3mL)。反應(yīng)混合物在0℃下攪拌30分鐘,通過TLC(展開劑比例為3:1的己烷:乙酸乙酯)監(jiān)測反應(yīng)進(jìn)程,確認(rèn)原料完全消耗。反應(yīng)完成后,向混合物中加入水,用二氯甲烷進(jìn)行萃取。合并有機(jī)層,用無水硫酸鈉干燥,過濾后減壓濃縮,得到6.0g綠色油狀粗產(chǎn)物。粗產(chǎn)物通過硅膠柱色譜純化,采用逐步梯度洗脫(洗脫劑比例從6:1至3:1的己烷:乙酸乙酯),得到4-溴-2-氟苯乙酮(5.0g,收率86%)。產(chǎn)物經(jīng)質(zhì)譜(ESI)確認(rèn),m/z 218 [M + H]+。
