以2-氯-3-甲基-5-硝基吡啶為原料合成2-氯-3-甲基-5-氨基吡啶的一般步驟如下:在2小時內(nèi),將鐵粉(9.75 g,0.174 mol,Sigma-Aldrich)分批加入攪拌中的2-氯-3-甲基-5-硝基吡啶(10 g,0.058 mol,Combi)的乙酸/水(29 mL:88 mL)溶液中。反應(yīng)3小時后,將混合物通過硅藻土過濾,濾餅用乙酸乙酯洗滌。分離有機(jī)層后,水層用乙酸乙酯萃取。合并的有機(jī)層依次用碳酸氫鈉水溶液和鹽水洗滌,并經(jīng)無水硫酸鈉干燥。減壓濃縮除去溶劑,得到6-氯-5-甲基吡啶-3-胺,為棕色固體(8.0 g,收率97%)。質(zhì)譜(MS)m/z = 142.03 [M + H]+。1H-NMR(300 MHz, DMSO-d6)δ: 7.54(d, J = 30 Hz, 1H),6.91-6.90(dd, J = 0.6 Hz和2.7 Hz, 1H),5.39(s, 2H),2.17(s, 3H)。
