5-氟吲哚(1.0 g,7.40 mmol)溶于1 mL乙酸中,將該溶液緩慢滴加到預(yù)先冷卻至0℃的二甲胺(567 mg,1.42 mL,12.58 mmol,40%水溶液)和甲醛(333 mg,0.9 mL,11.09 mmol,37%水溶液)的混合溶液中,反應(yīng)在氬氣保護(hù)下進(jìn)行。反應(yīng)混合物在不冷卻的條件下攪拌3小時(shí)。反應(yīng)完成后,將混合物倒入20 mL冰水中,并用堿調(diào)節(jié)pH至11。隨后,用乙醚萃取水相中的有機(jī)產(chǎn)物,合并有機(jī)相并用無水硫酸鎂干燥。減壓蒸發(fā)除去溶劑后,通過硅膠柱色譜法純化粗產(chǎn)物,洗脫劑為氯仿/甲醇/氨水(90:10:1至40:10:1梯度洗脫),得到目標(biāo)化合物1-(5-氟-1H-吲哚-3-基)-N,N-二甲基甲胺(1.33 g,6.92 mmol,收率93%),為米色固體。產(chǎn)物結(jié)構(gòu)經(jīng)1H NMR和13C NMR確認(rèn):1H NMR (400 MHz, CDCl3) δ 8.47 (s, 1H), 7.36-7.31 (m, 1H), 7.25-7.20 (m, 1H), 7.12 (d, J = 2.6 Hz, 1H), 6.95-6.88 (m, 1H), 3.58 (s, 2H), 2.28 (s, 6H); 13C NMR (100 MHz, CDCl3) δ 157.5 (d, J = 234.4 Hz), 132.4, 128.0 (d, J = 9.9 Hz), 125.1, 113.2 (d, J = 4.9 Hz), 111.3 (d, J = 9.7 Hz), 110.0 (d, J = 26.4 Hz), 103.87 (d, J = 23.5 Hz), 54.1, 44.9。
