在60 mL干燥的蛋形燒瓶中加入6-溴-2-萘甲酸(2.4996 g,10.0 mmol),隨后加入20 mL無水甲醇使其溶解。緩慢滴加1 mL濃硫酸至反應(yīng)體系中,然后將混合物回流過夜。通過TLC監(jiān)測(cè)反應(yīng)進(jìn)度,直至原料完全轉(zhuǎn)化。反應(yīng)完成后,停止加熱,待體系冷卻至室溫。用飽和碳酸鈉水溶液淬滅反應(yīng),調(diào)節(jié)反應(yīng)體系至中性。用乙酸乙酯萃取反應(yīng)混合物,有機(jī)相分別用飽和碳酸鈉水溶液、水和飽和氯化鈉水溶液各洗滌一次。有機(jī)層經(jīng)無水硫酸鈉干燥后,減壓濃縮,得到6-溴-2-萘甲酸甲酯(S7,2.63 g,收率100%)為白色固體。
