在100 mL圓底燒瓶中加入6-溴煙酸(540 mg,2.7 mmol)、二氯甲烷(20 mL)和N,N-二甲基甲酰胺(1滴)。隨后緩慢加入草酰氯(1 mL),觀察到劇烈氣體逸出。反應(yīng)混合物在室溫下攪拌1小時(shí),之后通過旋轉(zhuǎn)蒸發(fā)儀減壓除去溶劑。殘余物用甲醇(MeOH)處理,再次減壓除去溶劑。將殘余物溶解于二氯甲烷中,并用飽和碳酸氫鈉(NaHCO3)溶液洗滌。有機(jī)層用無水硫酸鈉(Na2SO4)干燥,過濾后減壓濃縮,得到6-溴煙酸甲酯369 mg,收率55%。產(chǎn)物經(jīng)1H NMR(500 MHz)表征:δ 9.00(dd,J = 2.0, 0.5 Hz,1H),8.25(dd,J = 8.5, 2.5 Hz,1H),7.42(dd,J = 8.5, 0.5 Hz,1H),3.96(s,3H)。
