(1) 2-氯煙酸甲酯的制備:將15.0 g (95.2 mmol) 2-氯煙酸溶解于150 mL二氯甲烷中,緩慢滴加8.37 mL (94.5 mmol) 草酰氯。隨后,向混合物中逐滴加入N,N-二甲基甲酰胺,并在室溫下攪拌反應(yīng)混合物3小時。反應(yīng)完成后,在冰浴冷卻條件下,依次滴加40.1 mL (286 mmol) 三乙胺和37 mL (914 mmol) 甲醇至反應(yīng)混合物中,繼續(xù)在冰浴冷卻下攪拌30分鐘。反應(yīng)混合物經(jīng)減壓濃縮后,向殘余物中加入碳酸氫鈉水溶液進(jìn)行中和。用乙醚萃取水相,合并有機相,用水洗滌,無水硫酸鈉干燥,過濾后濃縮。殘余物通過柱層析法純化(洗脫劑:乙酸乙酯/正己烷=1:4),得到13.9 g目標(biāo)產(chǎn)物2-氯煙酸甲酯,為淡黃色液體,收率86%。1H-NMR (CDCl3/TMS, δ ppm): 3.97 (3H, s), 7.33 (1H, dd), 8.17 (1H, dd), 8.53 (1H, dd)。
