嘧啶-2-羧酸(11)的合成:在攪拌條件下,將嘧啶-2-甲腈(10)(201 mg,1.914 mmol)溶于水(5 mL)中,隨后加入KOH(214.3 mg,3.83 mmol)。將反應(yīng)混合物加熱回流3小時(shí)。通過(guò)薄層色譜(TLC)監(jiān)測(cè)反應(yīng)進(jìn)度,確認(rèn)原料完全消耗后,將反應(yīng)混合物緩慢冷卻至室溫。用2N HCl溶液中和反應(yīng)混合物,隨后通過(guò)旋轉(zhuǎn)蒸發(fā)除去水相,得到粗產(chǎn)物。用乙酸乙酯(EtOAc)萃取粗產(chǎn)物,合并有機(jī)相并通過(guò)硅藻土墊過(guò)濾。濾液經(jīng)減壓濃縮后,得到嘧啶-2-羧酸(11)(84 mg,收率35.4%),該產(chǎn)物無(wú)需進(jìn)一步純化即可用于后續(xù)反應(yīng)。TLC條件:80% EtOAc/己烷(Rf:0.05)。1H NMR(400 MHz,CD3OD-d4):δ 8.83(br s,2H),7.47(t,J = 4.8 Hz,1H)。
