以2,5-二溴吡啶和N,N-二甲基甲酰胺(DMF)為原料,按照通用方法C合成5-溴-2-吡啶甲醛。具體操作如下:將2,5-二溴吡啶(237 mg, 1 mmol)與DMF(115 μL, 1.5 mmol)混合反應(yīng)。反應(yīng)完成后,通過(guò)快速柱色譜法(洗脫劑:己烷/乙酸乙酯=10:1)純化粗產(chǎn)物,得到目標(biāo)化合物5-溴-2-吡啶甲醛(27b),為褐色固體(149 mg, 產(chǎn)率80%)。薄層色譜(TLC)分析顯示Rf值為0.45(展開(kāi)劑:己烷/乙酸乙酯=8:1)。核磁共振氫譜(1H NMR, 400 MHz, CDCl3)數(shù)據(jù)如下:δ 10.03 (1H, s), 8.85 (1H, dd, J = 0.8, 2.0 Hz), 8.02 (1H, ddd, J = 0.8, 2.0, 8.0 Hz), 7.85 (1H, dd, J = 0.8, 8.0 Hz)。核磁共振碳譜(13C NMR, 100 MHz, CDCl3)數(shù)據(jù)如下:δ 192.4, 151.7, 151.3, 140.0, 126.3, 122.8。所得光譜數(shù)據(jù)與文獻(xiàn)報(bào)道一致。
