以甲醇和3-甲氧基-4-羥基苯基丙烯酸為原料合成(E)-3-(4-羥基-3-甲氧基苯基)丙烯酸甲酯的一般步驟:將阿魏酸(1.94 g,10 mmol)溶解于無水甲醇(3.20 g,100 mmol)中,加入0.25 mL濃硫酸作為催化劑。在攪拌下回流反應(yīng),通過薄層色譜(TLC)監(jiān)測反應(yīng)進程。待原料點消失后,停止反應(yīng)并冷卻至室溫。將反應(yīng)混合物倒入冰水體系中,用10%過量的碳酸鈉溶液中和剩余的酸。隨后,用飽和鹽水洗滌反應(yīng)混合物,并用乙醚進行萃取。分離有機相后,合并有機層并用無水硫酸鈉干燥。過濾除去干燥劑后,減壓蒸除溶劑得到粗產(chǎn)物。粗產(chǎn)物用正己烷洗脫,進一步通過重結(jié)晶純化,得到白色固體狀的(E)-3-(4-羥基-3-甲氧基苯基)丙烯酸甲酯,質(zhì)量為1.42 g(理論質(zhì)量為2.08 g),收率為68.3%。
