以喹啉-2-羧酸甲酯為原料合成2-羥甲基喹啉的一般步驟:在室溫下,將喹啉-2-羧酸甲酯(5g,26.8mmol)的四氫呋喃(THF,30mL)溶液緩慢加入至含有硼氫化鈉(NaBH4,710mg,18.8mmol)的四氫呋喃(THF,20mL)溶液中。將反應(yīng)混合物于35℃下攪拌30分鐘。隨后,在相同溫度下依次加入甲醇(2.5mL)、溫水(30mL)和乙酸乙酯(20mL)。分離有機(jī)層,并用去離子水(2×30mL)洗滌。有機(jī)相經(jīng)無水硫酸鎂干燥后,過濾并減壓濃縮。粗產(chǎn)物通過硅膠柱色譜法(洗脫劑:乙酸乙酯/甲基環(huán)己烷=60/40)進(jìn)行純化,得到白色固體產(chǎn)物,收率73%。產(chǎn)物熔點(diǎn)為135℃(分解)。核磁共振氫譜(1H NMR,300MHz,CDCl3)顯示:δ 4.68(1H,s,OH),4.95(2H,s,CH2),7.31(1H,m,芳香CH),7.55(1H,m,芳香CH),7.71(1H,m,芳香CH),7.81(1H,m,芳香CH),8.10(2H,m,芳香CH)。核磁共振碳譜(13C NMR,75MHz,CDCl3)顯示:δ 64.25, 118.43, 126.35, 127.55, 127.69, 128.57, 129.81, 136.86, 146.72, 159.19。元素分析計(jì)算值(C10H9NO):C,75.45;H,5.70;N,8.80。實(shí)測(cè)值:C,75.13;H,5.56;N,8.75。
