將2'-脫氧-2'-氟尿苷(3.00 g,12.2 mmol)溶解于無水吡啶中,重復進行減壓濃縮操作3次以實現(xiàn)脫水共沸蒸餾。隨后,在氬氣保護下,將反應混合物溶解于無水吡啶(120 mL)中,加入4,4'-二甲氧基三苯甲基氯(4.55 g,13.4 mmol),室溫攪拌反應3小時。反應完成后,向反應混合物中加入乙酸乙酯(150 mL)和水(60 mL),進行分層。有機層依次用5%碳酸氫鈉水溶液(20 mL×3)、水(20 mL)和飽和食鹽水(20 mL)洗滌,隨后用無水硫酸鈉干燥。過濾后,蒸發(fā)去除溶劑,殘余物通過硅膠柱色譜純化(洗脫劑:己烷/乙酸乙酯=50:50至0:100(v/v),含1%三乙胺)。收集含目標產(chǎn)物的餾分,濃縮后得到1-((2R,3R,4R,5R)-5-((雙(4-甲氧基苯基)(苯基)甲氧基)甲基)-3-氟-4-羥基四氫呋喃-2-基)嘧啶-2,4(1H,3H)-二酮(8.48 g,定量收率)。
