2 Preparation of 2-amino-4-methyl-5-(4-nitrophenyl)thiophene-3-carboxylic acid ethyl ester
Dissolve 1 kg of 1-(4-nitrophenyl)propan-2-one in 4L of ethanol, add 685 g of ethyl cyanoacetate and 195 g of sulfur, and stir.The temperature was raised to 40°C, 200 g of n-butylamine was added, and the temperature was raised to 50°C and stirred for 2 hours.Cool down to about 10°C and stir for 1 hour.After filtration, the filter cake was rinsed with 2L ethanol to obtain 1.37 kg of product, with a purity of 99% and a yield of 85.0%.
