Synthesis | The general procedure for the synthesis of 2-(4-bromophenyl)-5-phenylthiophene from p-bromoiodobenzene and (5-phenylthiophen-2-yl)boronic acid is as follows: 20.4 g (100 mmol, 1.0 eq.) of (5-phenylthiophen-2-yl)boronic acid and 31.1 g (110 mmol, 1.1 eq.) of p-bromoiodobenzene were added to a 2 L three-necked flask containing 1000 mL toluene in a 2 L three-necked flask containing 1000 mL of toluene. Subsequently, 100 mL of ethanol was added and stirred until complete dissolution. The reaction system was purged with nitrogen for 15 minutes to remove oxygen, then 150 mL of a 2M aqueous solution containing 41.5 g (300 mmol, 3.0 eq.) of K2CO3 was added. Finally, 2.3 g (2 mol%) of Pd(PPh3)4 was added as a catalyst. The reaction mixture was heated to 110 °C and the reaction was stirred at this temperature overnight. Upon completion of the reaction, activated carbon was added for adsorption, followed by filtration to remove the activated carbon. The filtrate was concentrated under reduced pressure to remove the solvent, and the crude product obtained was dried and recrystallized from toluene and ethanol to give 26.2 g of Intermediate-9 in 83% yield. | References | [1] Patent: CN108101896, 2018, A.
