2-氯-5-甲氧基嘧啶-4-胺的合成步驟如下:在氬氣保護(hù)下,將2,4-二氯-5-甲氧基嘧啶(10g,56.17mmol)溶于1,4-二惡烷(20mL)中,室溫攪拌。隨后,向該溶液中緩慢加入氫氧化銨(25.5mL)。將反應(yīng)混合物轉(zhuǎn)移至密封管中,加熱至100℃并維持21小時。反應(yīng)進(jìn)程通過薄層色譜(TLC)監(jiān)測。反應(yīng)完成后,在減壓條件下蒸除揮發(fā)性組分。粗產(chǎn)物通過硅膠柱色譜法純化,洗脫劑為40%乙酸乙酯/己烷,得到2-氯-5-甲氧基嘧啶-4-胺(8.9g,收率99%),為灰白色固體。產(chǎn)物結(jié)構(gòu)經(jīng)1H-NMR(DMSO-d6, 500MHz)確認(rèn):δ7.68(s, 1H),3.81(s, 3H);LC-MS(柱:X-Bridge C-18,50×3.0mm,3.5μm;流動相:0.05%三氟乙酸水溶液/乙腈;流速:0.80mL/min)顯示m/z 160([M+H]+),保留時間2.18分鐘;TLC(展開劑:50%乙酸乙酯/己烷)顯示Rf值為0.3。
