在氮氣氛圍下,于25℃條件下,向4-溴苯甲醛(2.0g,0.010mol)的N,N-二甲基甲酰胺(DMF,20mL)溶液中依次加入1H-吡唑(0.668g,0.0098mol)、碘化銅(0.185g,0.0009mol)、L-脯氨酸(0.224g,0.0019mol)及碳酸銫(6.4g,0.0196mol)。將反應(yīng)混合物加熱至120℃并攪拌反應(yīng)20小時。反應(yīng)完成后,冷卻至室溫,通過硅藻土墊過濾,并用乙酸乙酯(50mL)洗滌濾餅。合并濾液,減壓濃縮除去溶劑,得到粗產(chǎn)物。粗產(chǎn)物通過快速柱色譜法純化,洗脫劑為乙酸乙酯/正己烷(40:60,v/v),得到目標化合物4-(吡唑-1-基)苯甲醛。產(chǎn)量:1.0g。核磁共振氫譜(DMSO-d6, 400MHz)δppm:6.62(s, 1H),7.84(s, 1H),8.01-8.03(d, J = 8.4Hz, 2H),8.07(d, J = 8.4Hz, 2H),8.67(s, 1H),10.00(s, 1H)。質(zhì)譜(m/z):173.1([M + H]+)。
