以呋喃并[3,2-d]嘧啶-2,4-二醇(7J)(1.52 g,10 mmol)為起始原料,加入N,N-二甲基苯胺(1 mL,8 mmol)和三氯氧磷(0.9 mL,9.65 mmol),在130°C下反應(yīng)3小時。反應(yīng)完成后,將混合物冷卻至室溫,并小心地用冰水淬滅。通過過濾收集生成的固體產(chǎn)物,用水洗滌,并在真空下干燥,得到2,4-二氯呋喃并[3,2-d]嘧啶(7K)(1.02 g,產(chǎn)率54%),為淺棕色固體。產(chǎn)物的熔點為107.3°C。核磁共振氫譜(300 MHz,CDCl3)數(shù)據(jù)如下:δ 8.08(d,J = 2.2 Hz,1H),7.02(d,J = 2.2 Hz,1H)。
