向1000 mL三頸燒瓶中加入化合物1t(54.0 g,0.25 mol)和丙酮(500 mL)。隨后加入QAS-2(11.4 g,25.0 mmol)。將反應(yīng)混合物冷卻至0-5℃。在10分鐘內(nèi)分批加入固體KMnO4(47.5 g,0.3 mol)。保持該溫度下攪拌反應(yīng)混合物2小時(shí),通過(guò)TLC監(jiān)測(cè)反應(yīng)直至原料1t完全消耗。反應(yīng)完成后,一次性加入NaHSO3(25.0 g)的水(50 mL)溶液以淬滅反應(yīng)。繼續(xù)攪拌15分鐘,然后通過(guò)硅藻土過(guò)濾,用丙酮(3×100 mL)徹底洗滌濾餅。合并濾液,減壓濃縮以除去丙酮。殘余物用EtOAc(3×150 mL)萃取。合并有機(jī)相,依次用飽和NaHCO3溶液和鹽水(3×50 mL)洗滌,無(wú)水Na2SO4干燥,過(guò)濾后減壓濃縮至干,得到無(wú)色油狀物(57.5 g,收率93%)。該油狀物通過(guò)石油醚(200 mL)重結(jié)晶,得到白色固體產(chǎn)物,產(chǎn)量51.3 g(收率83%),熔點(diǎn)75-76℃。1H NMR(400 MHz,CDCl3)數(shù)據(jù)如下:δ= 4.20-4.31(m,3H),4.09(m,2H),3.93(d,J = 8.0 Hz,1H),2.52(br,2H),1.31-1.49(m,12H)。
