以N-BOC-3-哌啶甲酰胺為原料合成3-氰基哌啶-1-羧酸叔丁酯(3)的一般步驟:向叔丁基-3-氨基甲?;哙?1-羧酸酯(2)(1.0g,4mmol)的無(wú)水二氯甲烷(20mL)溶液中加入三乙胺(3.1g,31.0mmol),隨后緩慢滴加三氟乙酸酐(4.1g,20.0mmol)。將反應(yīng)混合物逐漸升溫至室溫并持續(xù)攪拌16小時(shí)。反應(yīng)完成后,將混合物在二氯甲烷(50mL)和水(50mL)之間分配。有機(jī)相用飽和碳酸氫鈉溶液(100mL)洗滌,隨后用無(wú)水硫酸鈉干燥。減壓濃縮有機(jī)相,得到粘稠液體產(chǎn)物3(800mg,收率87%)。1H NMR(400MHz,CDCl3)δ1.49(s,9H),1.77(m,2H),1.97(brs,1H),2.66(m,1H),3.35-3.84(brs,4H)。
