以烏洛托品和2,4-二氯酚為原料合成3,5-二氯水楊醛的一般步驟如下:首先,將2,4-二氯苯酚(10.0g,61.3mmol)和六亞甲基四胺(17.2g,122.6mmol)溶解于80mL甲磺酸中,反應(yīng)混合物在100℃下加熱攪拌1.5小時(shí)。反應(yīng)完成后,用乙酸乙酯稀釋反應(yīng)混合物,依次用水和飽和碳酸氫鈉溶液洗滌有機(jī)層,然后用無水硫酸鎂干燥。減壓蒸發(fā)除去溶劑,得到粗產(chǎn)物。粗產(chǎn)物通過柱色譜法純化,采用乙酸乙酯/己烷梯度洗脫,得到目標(biāo)化合物3,5-二氯-2-羥基苯甲醛(INT-01)為黃色油狀物(7.5g,收率64%)。產(chǎn)物結(jié)構(gòu)經(jīng)1H NMR(400MHz,CDCl3)確認(rèn):δ11.39(s,1H),7.64(s,1H),7.52(s,1H)。液相色譜(LC)分析條件:C-18柱,5分鐘至95%乙腈/水梯度洗脫,流速1.7mL/min,檢測波長254nm,柱溫23℃,保留時(shí)間(tr)=3.95分鐘。
