一般步驟:在冰浴冷卻條件下,將NaH(60%分散于礦物油中,40.00 mmol,0.96 g)分批加入攪拌的3-吲哚甲醛(20.00 mmol,2.90 g)的無水THF(50 mL)溶液中。緩慢升溫至室溫后,繼續(xù)攪拌30分鐘。隨后,滴加苯磺酰氯(24.00 mmol,4.24 g)的無水THF溶液。通過TLC監(jiān)測反應進程,待原料3-吲哚甲醛完全消失后,減壓蒸發(fā)除去溶劑。將反應混合物用冰水(300 mL)稀釋,過濾收集固體產(chǎn)物。用丙酮/石油醚(60-90℃)重結晶,得到目標產(chǎn)物1-(苯磺酰)-3-吲哚甲醛,產(chǎn)率97%,熔點151-153℃。1H NMR(600 MHz,CDCl3):δ 7.37(t,J = 7.2 Hz,1H),7.42(dd,J = 11.4, 4.2 Hz,1H),7.52(t,J = 7.8 Hz,2H),7.62(t,J = 7.2 Hz,1H),7.97(t,J = 7.8 Hz,3H),8.20-8.31(m,2H),10.11(s,1H)。HRMS(MALDI):m/z 286.0515。計算值C15H11NO3S:[M + H]+ 286.0538。
