以化合物(CAS: 198641-56-4)為原料合成(2R,4R)-4-甲基-2-哌啶甲酸乙酯的一般步驟:將(1'S,2R,4R)-6(2.5 g,9 mmol)溶解于乙醇(120 mL)中,加入10% Pd(OH)2/C(0.27 g)。在室溫及常壓下進行氫化反應(yīng)。反應(yīng)進程通過TLC(展開劑:氯仿/甲醇,9:1)監(jiān)測。反應(yīng)4小時后,通過硅藻土墊過濾去除催化劑。減壓濃縮濾液,得到粗產(chǎn)物。粗產(chǎn)物通過硅膠柱色譜(硅膠與樣品質(zhì)量比為1:20)進行純化,以二氯甲烷/甲醇(98:2)為洗脫劑,得到純的(2R,4R)-4-甲基-2-哌啶甲酸乙酯(0.7 g,收率45%)。產(chǎn)物經(jīng)1H NMR(CDCl3)表征:δ 0.96(d,3H,J = 6.50 Hz,CH3-4);1.15(m,1H,H-5a);1.32(t,3H,J = 7.2 Hz,CH3CH2);1.42-1.54(m,1H,H-3);1.5-1.69(m,2H,H-4,H-5b);2.00-2.11(m,2H,H-3b和NH);2.87(m,2H,H-6);3.65(m,1H,H-2);4.21(q,2H,J = 7.2 Hz,CH2CH3)。比旋光度[α]20D = -22(c 5,乙醇)(對映體過量值e.e. = +24)。
