以二碳酸二叔丁酯和反式-4-羥基-L-脯氨酸甲酯鹽酸鹽為原料合成N-Boc-反式-4-羥基-L-脯氨酸甲酯的一般步驟如下:將步驟I中得到的反式-4-羥基-L-脯氨酸甲酯鹽酸鹽(14.5g,0.1mol)懸浮于CH2Cl2(400mL)中,冷卻至0℃,攪拌下依次加入Et3N(28mL,0.2mol)、DMAP(0.61g,5mmol)和Boc酐(27.5mL,0.12mol)。反應(yīng)混合物逐漸升溫至室溫并繼續(xù)攪拌2小時。反應(yīng)完成后,減壓濃縮除去溶劑,向殘留固體中加入乙醚,過濾收集固體并用乙醚充分洗滌。合并濾液,減壓濃縮。將殘余物溶解于CH2Cl2中,依次用飽和NaHCO3水溶液和飽和NaCl水溶液洗滌,有機相用無水Na2SO4干燥。減壓蒸除溶劑,得到淺黃色油狀物,經(jīng)高真空處理固化。所得固體用己烷多次研磨,高真空干燥,得到1-叔丁基-2-甲基(2S,4R)-4-羥基吡咯烷-1,2-二甲酸酯,為白色固體(24.5g,收率100%)。產(chǎn)物表征數(shù)據(jù)如下:(M+1) 245; 1H NMR (CDCl3, 300MHz) δ 4.54-4.37 (m, 2H), 3.75 (s, 3H), 3.70-3.40 (m, 2H), 2.48-2.21 (m, 2H), 2.13-2.0 (m, 1H), 1.46 (s, 3H), 1.41 (s, 6H)。
