以1H-吲唑-6-羧酸甲酯為原料合成1H-吲唑-6-羧酸的一般步驟:向攪拌的1H-吲唑-6-羧酸甲酯(1.0g,5.676mmol,1當(dāng)量)在四氫呋喃/乙醇/水(1:1:1,10mL)的混合溶劑中的溶液中加入氫氧化鋰(476mg,11.353mmol,2當(dāng)量)。反應(yīng)混合物在室溫下攪拌16小時(shí)。反應(yīng)完成后,蒸發(fā)除去溶劑,殘余物用10mL水稀釋?zhuān)⒂?N鹽酸調(diào)節(jié)pH至4-5,得到固體沉淀。通過(guò)過(guò)濾收集沉淀,干燥后得到1H-吲唑-6-羧酸(900mg,收率97%),為灰白色固體。TLC條件:甲醇/二氯甲烷(1:9),Rf值為0.1。1H NMR(300MHz,DMSO-d6):δ13.36(s,1H),13.04(s,1H),8.24-8.08(m,2H),7.85(dd,J=8.5,0.9Hz,1H),7.67(dd,J=8.4,1.3Hz,1H)。
