以4-(3-氟苯基)丁酸為原料合成6-氟-1-四氫萘酮的一般步驟:將4-(3-氟苯基)丁酸(1.92g,10.5mmol)與多磷酸(2g)的混合物在氮?dú)獗Wo(hù)下于110°C加熱2小時(shí)。反應(yīng)完成后,冷卻至室溫,用水淬滅反應(yīng),用乙醚稀釋反應(yīng)混合物。隨后,用飽和碳酸氫鈉水溶液洗滌(2次),有機(jī)相用無(wú)水硫酸鈉干燥,濃縮后得到6-氟-1-四氫萘酮(1.51g,收率87%)。產(chǎn)物經(jīng)NMR(400MHz,CDCl3)表征:δ2.16(m,2H),2.64(t,2H),2.97(t,2H),6.92(dd,1H),6.99(dt,1H),8.04(dd,1H)。
