以D,L-苯丙氨酸(50g,3.03mol)為原料,制備其混懸液。在劇烈攪拌下,向混懸液中加入濃鹽酸(325mL)和37%甲醛溶液(110mL),加熱至溫和回流狀態(tài),維持30分鐘。隨后,額外加入甲醛(50mL)和濃鹽酸(110mL),繼續(xù)加熱反應(yīng)4小時(shí)。反應(yīng)完成后,將反應(yīng)混合物冷卻至室溫,通過過濾收集固體產(chǎn)物。用甲醇(30mL)洗滌固體,干燥后得到目標(biāo)化合物3-羧基-1,2,3,4-四氫異喹啉,產(chǎn)量64.09g(產(chǎn)率98.9%),熔點(diǎn)為295-298℃(分解溫度>326℃)(文獻(xiàn)參考:Julian PL, J. Chem. Soc. 1948, 70, 180-183),為對(duì)映體混合物。其核磁共振氫譜(DMSO-d6)數(shù)據(jù)如下:δ 3.10-3.18(1H,m,CH),3.28-3.34(1H,m,CH),4.27(2H,s,CH2),4.39(1H,m,CH),7.25-7.27(4H,m,4*ArH),9.89(1H,brs,可交換,NH),10.06(1H,brs,可交換,OH)。質(zhì)譜數(shù)據(jù):m/z 177.0(M+H)+。
