一般步驟:將(8R,9S,10R,13S,14S,17R)-17-乙酰基-17-羥基-10,13-二甲基-6,7,8,9,10,11,12,13,14,15,16,17-十二氫-1H-環(huán)戊烷并[a]菲-3(2H)-酮(17α-羥基黃體酮,20g)、己酸(40ml)、吡啶(16ml)、對(duì)甲苯磺酸(1.6g)和甲苯(300ml)加入500ml三頸燒瓶中。將反應(yīng)混合物加熱至110~120℃,維持3小時(shí)。反應(yīng)過(guò)程中通過(guò)薄層色譜(TLC)監(jiān)測(cè)反應(yīng)進(jìn)度。反應(yīng)完成后,對(duì)反應(yīng)混合物進(jìn)行減壓濃縮,以去除甲苯、吡啶和未反應(yīng)的己酸。濃縮后,向三頸燒瓶中加入100ml乙醇和3ml濃鹽酸,加熱回流2小時(shí)。再次通過(guò)TLC監(jiān)測(cè)反應(yīng),確保17α-羥基黃體酮完全轉(zhuǎn)化為(8R,9S,10R,13S,14S,17R)-17-乙?;?10,13-二甲基-3-氧代-2,3,6,7,8,9,10,11,12,13,14,15,16,17-十四氫-1H-環(huán)戊并[a]菲-17-基己酸酯(己酸黃體酮)。反應(yīng)完成后,將混合物冷卻至5℃以下,過(guò)濾得到己酸黃體酮粗品(24g,粗產(chǎn)率120%)。粗品通過(guò)乙醇重結(jié)晶純化,得到純品19.8g,純化收率82.5%,總收率99.0%。
