以乙醇和丙炔酸為原料合成丙炔酸乙酯的一般步驟如下:將丙炔酸(310μL,5mmol;Alfa Aesar,Ward Hill,MA;目錄號A13245)溶于13mL乙醇中,形成無色溶液,隨后向該溶液中緩慢加入濃硫酸(139μL,2.5mmol)。將反應(yīng)混合物加熱至回流狀態(tài),持續(xù)反應(yīng)24小時。反應(yīng)完成后,用13mL水淬滅反應(yīng),隨后用二氯甲烷(10mL;三次)進(jìn)行萃取。合并有機(jī)相,依次用水(10mL;三次)和鹽水(10mL)洗滌,然后用無水Na2SO4(Thermo Fisher Scientific,Pittsburgh,PA;目錄號S421-3)干燥。減壓蒸餾去除溶劑,得到淺黃色油狀產(chǎn)物丙炔酸乙酯(177mg,收率36%)。產(chǎn)物經(jīng)1H NMR(400MHz,CDCl3,δ)表征:4.26(q,3JH-H = 7.1Hz,CH2,2H),2.92(s,C≡CH,1H),1.33(t,3JH-H = 7.2Hz,CH3,3H)。
