以3,4-二氫-2H-吡喃和2-丙炔-1-醇為原料合成O-THP炔丙醇(化合物18)的一般步驟:化合物18根據(jù)文獻(xiàn)程序制備。向炔丙醇17(2.6 mL,44.6 mmol)和對甲苯磺酸一水合物(0.09 g,0.5 mmol)在二氯甲烷(45 mL)中的溶液于0℃下滴加3,4-二氫-2H-吡喃(4.3 mL,46.8 mmol)。反應(yīng)混合物在0℃下攪拌5分鐘后,緩慢升溫至室溫并繼續(xù)攪拌1小時。隨后,反應(yīng)混合物用飽和碳酸氫鈉溶液(30 mL)洗滌,水層用二氯甲烷(45 mL)萃取。合并有機(jī)層,經(jīng)無水硫酸鎂干燥,過濾后減壓濃縮,得到化合物18(6.1 g,收率97.2%),為黃色油狀物。薄層色譜(TLC)Rf = 0.30(正己烷/乙醚,95:5)。1H NMR(400 MHz,CDCl3):δ = 1.50-1.67(m,4H),1.70-1.89(m,2H),2.41(t,J = 2.4 Hz,1H),3.52-3.57(m,1H),3.82-3.87(m,1H),4.27(ddd,J = 2.4 Hz,11.3 Hz,15.8 Hz,2H),4.83(t,J = 3.4 Hz,1H)。13C NMR(100 MHz,CDCl3):δ = 19.0, 25.3, 30.2, 53.9, 61.9, 74.0, 79.7, 96.8。質(zhì)譜(MS-CI):m/z計算值C8H12O2 [M + H]+,141;實測值:141。此化合物的合成方法已有文獻(xiàn)報道。
