以3-溴氰芐為原料合成3-溴苯乙胺的一般步驟:將LiAlH4(3.04 g,80 mmol)的無水THF(100 mL)懸浮液冷卻至-5°C。緩慢滴加濃H2SO4(3.9 g,40 mmol),并將所得混合物在-5°C下攪拌1小時。隨后,緩慢滴加3-溴苯乙腈(9.80 g,50 mmol)的THF(5 mL)溶液,滴加完畢后將反應體系緩慢升溫至室溫。反應在室溫下繼續(xù)攪拌1小時,然后冷卻至0°C,并通過加入1:1 THF:H2O混合物(12.4 mL)淬滅反應。加入Et2O(50 mL),隨后加入3.6 M NaOH溶液(24.4 mL)。將混合物通過硅藻土過濾,并用額外的Et2O充分洗滌固體。合并有機相,用Na2SO4干燥,過濾后真空濃縮,得到3-溴苯乙胺(9.7 g,收率97%)。粗產(chǎn)物直接用于后續(xù)反應。1H NMR(400 MHz,CDCl3)δ 7.38-7.30(m,2H),7.20-7.10(m,2H),2.96(t,J = 7.2 Hz,2H),2.72(t,J = 7.2 Hz,2H),1.35(br s,2H)。MS(ESI)m/z:計算值:199;實測值:200/202(M+·)。
