步驟1:1-溴-4-乙氧基苯的合成:在室溫下,將碳酸鉀(118 g)加入至4-溴苯酚(50 g)的丙酮(250 mL)溶液中,攪拌30分鐘。隨后,緩慢滴加碘乙烷(67.6 g)。將反應(yīng)混合物加熱回流10小時(shí),并通過薄層色譜(TLC)監(jiān)測(cè)反應(yīng)進(jìn)程。反應(yīng)完成后,過濾混合物,使用旋轉(zhuǎn)蒸發(fā)器去除丙酮。將殘余物溶解于乙酸乙酯中,依次用水和飽和鹽水洗滌,經(jīng)無水硫酸鈉干燥后濃縮。通過硅膠柱色譜法,以乙酸乙酯和正己烷(1:7)為洗脫劑進(jìn)行純化,得到4-溴苯乙醚,為無色液體(57 g,產(chǎn)率98.27%)。1H NMR(CDCl3, 300 MHz):δ 7.35(d, J = 8.7 Hz, 2H),6.76(d, J = 8.7 Hz, 2H),3.99(q, J = 6.9 Hz, 2H),1.40(t, J = 6.9 Hz, 3H)。
