向150 mL圓底燒瓶中加入28.5 g (0.21 mol) 對甲氧基苯甲醛、21.8 g (0.21 mol) 丙二酸和32.3 g (0.42 mol) 乙酸銨,溶解于150 mL乙醇中。將反應(yīng)混合物加熱至75-80°C,在回流條件下攪拌反應(yīng)8小時。反應(yīng)完成后,將反應(yīng)混合物冷卻至室溫并繼續(xù)攪拌20小時。隨后,通過過濾收集白色固體,得到23.6 g 3-氨基-3-(4-甲氧基苯基)丙酸(外消旋混合物),產(chǎn)率為82.3%(基于對甲氧基苯甲醛)。產(chǎn)物為白色粉末,其物理性質(zhì)如下:1H-NMR (δ, ppm, D2O): 2.50 (dd, 1H, J = 17.1, 6.8 Hz), 2.65 (dd, 1H, J = 17.1, 7.8 Hz), 3.24 (s, 3H), 6.46 (s, 1H), 6.48 (s, 1H), 6.86 (s, 1H), 6.88 (s, 1H); 13C-NMR (δ, ppm, D2O): 37.1, 50.7, 55.1, 114.3, 126.9, 128.2, 159.1, 172.7; MS (EI) m/z: 195 (M+); MS (CI, i-C4H10) m/z: 196 (MH+).
