以N-甲基哌嗪和2-溴乙醇為原料合成1-(2-羥乙基)-4-甲基哌嗪的一般步驟如下:將2-溴乙醇(2.36 g,19 mmol)緩慢滴加到含有1-甲基哌嗪(1.26 g,13 mmol)和碳酸鉀(5.0 g,36 mmol)的無水乙醇(150 μL)混合物的反應(yīng)瓶中。反應(yīng)混合物在回流條件下加熱18小時。反應(yīng)完成后,使混合物冷卻至室溫,過濾以除去不溶性沉淀物。隨后,通過旋轉(zhuǎn)蒸發(fā)去除溶劑。殘余物用丙酮/二氯甲烷(體積比未指定)混合溶劑處理,再次過濾以除去不溶物,最后通過旋轉(zhuǎn)蒸發(fā)去除溶劑,得到1-(2-羥乙基)-4-甲基哌嗪(870 mg,產(chǎn)率48%),產(chǎn)物為淺棕色油狀物。產(chǎn)物經(jīng)1H NMR(CDCl3)確認(rèn):δ 2.18(s,3H);2.3-2.7(br m,8H);2.56(t,2H);3.61(t,2H)。MS-ESI顯示分子離子峰為145 [MH]+。
