以N,N-二甲基甲酰胺(DMF)和4,6-二羥基嘧啶為原料合成4,6-二氯-5-嘧啶甲醛的一般步驟:首先,在0℃下將DMF(64mL)與三氯氧磷(POCl3)(200mL)混合攪拌1小時。隨后,向該混合物中加入4,6-二羥基嘧啶(50.0g,446mmol),并在室溫下繼續(xù)攪拌0.5小時。接著,將所得非均相混合物加熱回流3小時。反應完成后,通過減壓蒸餾去除揮發(fā)性物質(zhì),將殘余物小心倒入冰水中。用乙醚對水相進行六次萃取,合并有機相。有機相依次用碳酸氫鈉(NaHCO3)水溶液和純水洗滌,無水硫酸鈉(Na2SO4)干燥。減壓濃縮干燥后的有機相,最后通過乙酸乙酯(EtOAc)-石油醚混合溶劑結(jié)晶,得到目標產(chǎn)物4,6-二氯-5-嘧啶甲醛(43.5g,收率55%)。產(chǎn)物經(jīng)LC-MS(ESI)分析,m/z 177 [M+H]+。
