一般步驟:在攪拌條件下,將草酰氯(0.3 mL)緩慢滴加至冷卻(冰?。┑腘,N-二甲基甲酰胺(DMF,3 mL)中。反應(yīng)混合物在0℃下持續(xù)攪拌1小時(shí)。隨后,將5-甲基吲哚(4 mmol)溶于DMF(1.5 mL)的溶液緩慢滴加入上述反應(yīng)混合物中。所得混合物于室溫下攪拌反應(yīng)5小時(shí)。反應(yīng)完成后,加入2N氫氧化鈉溶液(2 mL),并將混合物加熱至100℃維持10分鐘。反應(yīng)液冷卻后,用乙酸乙酯(3×50 mL)進(jìn)行萃取。合并有機(jī)相,依次用水和飽和鹽水洗滌。有機(jī)層經(jīng)無水硫酸鈉干燥后,減壓濃縮得到粗產(chǎn)物。粗產(chǎn)物通過快速柱色譜法純化,洗脫劑為乙酸乙酯/石油醚(3:1,v/v),最終得到純化的5-甲基-1H-吲哚-3-甲醛。
