以3-(2-噻吩甲?;?丙酸為原料合成4-(2-噻嗯基)丁酸的一般步驟如下:將4-氧代-4-(噻吩-2-基)丁酸(2.3g,12.48mmol)溶于乙二醇(30mL)中,依次加入水合肼(99%,2.2mL,45.9mmol)和氫氧化鉀顆粒(2.37g,42.4mmol)。將反應(yīng)混合物加熱至180℃,維持10小時。反應(yīng)完成后,冷卻至室溫,用水稀釋反應(yīng)混合物。水層用乙醚洗滌后,用6N鹽酸酸化,再用乙醚萃取。合并有機(jī)層,用無水硫酸鈉干燥,減壓濃縮。粗產(chǎn)物通過柱色譜法純化(硅膠60-120目,洗脫液為2%甲醇的二氯甲烷溶液),得到4-(噻吩-2-基)丁酸(1.8g,產(chǎn)率85%)。產(chǎn)物經(jīng)1H NMR(300MHz,DMSO-d6)和質(zhì)譜(ESI)確認(rèn):1H NMR δ 12.06(s,1H),7.31-7.29(m,1H),6.94-6.91(m,1H),6.84-6.82(m,1H),2.82-2.77(t,J=7.7Hz,2H),2.27-2.22(t,J=7.3Hz,2H),1.86-1.76(m,2H);MS(ESI)m/z: [M+H]+ 計(jì)算值170.04,實(shí)測值170.8。
