以2-氨基-6-氟苯并噻唑?yàn)樵虾铣?-氯-6-氟苯并噻唑的一般步驟:在圓底燒瓶中加入2-氨基-6-氟苯并噻唑(1.0017 g,5.96 mmol,Sigma-Aldrich Chemical Company, Inc.)、氯化銅(II)(1.201 g,8.93 mmol,Sigma-Aldrich Chemical Company, Inc.)和乙腈(ACN)中的亞硝酸叔丁酯(1.063 mL,8.93 mmol,Acros Organics)。將反應(yīng)混合物加熱至65℃,并在此溫度下保持2小時(shí)。反應(yīng)完成后,用1N鹽酸稀釋反應(yīng)混合物,并用乙酸乙酯(EtOAc)萃取。有機(jī)相依次用水和飽和氯化鈉溶液洗滌,經(jīng)無水硫酸鎂(MgSO4)干燥,過濾后減壓濃縮,得到目標(biāo)產(chǎn)物2-氯-6-氟苯并噻唑(0.9249 g,4.93 mmol,收率83%)。產(chǎn)物結(jié)構(gòu)經(jīng)1H NMR(400 MHz,DMSO-d6)確認(rèn):δ 7.45(td,J = 9.10, 2.74 Hz,1H),7.97-8.07(m,2H)。
