通用方法:5.1.2 7-甲氧基-7-氧代庚酸(84)的合成:在0℃下,將KOH(5.87g,104.65mmol)的甲醇(150ml)溶液緩慢滴加到庚二酸二甲酯(82,16.94g,90mmol)中。反應混合物在室溫下攪拌4小時。反應完成后,減壓除去溶劑,隨后加入乙醚(100ml)和水(200ml)進行萃取。分離有機相并濃縮,得到黃色油狀物82(5.08g,收率30%)。水相用濃鹽酸酸化至pH 3,并用乙醚(100ml×3)萃取。合并的有機層用鹽水(100ml×3)洗滌,經(jīng)硫酸鎂干燥后,減壓濃縮溶劑。產(chǎn)物通過硅膠柱色譜法純化,得到無色油狀物84(5.96g,收率38%)?;衔?5的合成采用相同的方法。5.1.2.1 8-甲氧基-8-氧代辛酸(85)的合成:收率44%。ESI-MS m/z:187.4 [M-H]-;1H NMR(DMSO-d6)δ1.21-1.32(m,4H),1.43-1.53(m,4H),2.18(t,J=7.2Hz,2H),2.29(t,J=7.2Hz,2H),3.58(s,3H),11.97(s,1H)。
