以5-(4-溴苯基)-3,4-二氫-2H-吡咯為原料合成2-(4-溴苯基)-吡咯烷的一般步驟:在-41℃下,將NaBH4(1.52g,40mmol)加入到5-(4-溴苯基)-3,4-二氫-2H-吡咯(4.48g,20mmol)溶于H2O/MeOH(30mL,v/v 1:4)的溶液中。攪拌反應(yīng)混合物4小時(shí)后,緩慢升溫至室溫。通過(guò)TLC監(jiān)測(cè)反應(yīng)完成后,加入2N HCl以淬滅未反應(yīng)的NaBH4。隨后,用去離子水和乙醚稀釋反應(yīng)混合物,分離有機(jī)層和水層。水層用乙醚進(jìn)一步萃取,合并有機(jī)相后用4M NaOH調(diào)節(jié)pH至12-13,并用乙酸乙酯洗滌。合并所有有機(jī)萃取液,用飽和食鹽水洗滌,無(wú)水Na2SO4干燥。減壓濃縮除去溶劑,得到粗產(chǎn)物2-(4-溴苯基)-吡咯烷,為黃色油狀物(3.8g,收率84%),該產(chǎn)物無(wú)需進(jìn)一步純化即可用于后續(xù)反應(yīng)。LC-MS(ESI)m/z:226(M + 1)+。
