以(1-甲基-4-哌啶基)[3-[2-(3-氯苯基)乙基]吡啶-2-基]甲酮鹽酸鹽為起始原料,合成8-氯-11-(1-甲基哌啶-4-亞甲基)-6,11-二氫-5H-苯并[5,6]環(huán)庚[1,2-b]吡啶的一般步驟如下:實施例2中,制備8-氯-6,11-二氫-1H-(N-甲基-4-哌啶亞基)-5H-苯并[5,6]環(huán)庚[1,2-b]吡啶(XII)。具體操作為:將化合物XIII(100g)按照美國專利No.4,659,716中所述的標準方法制備,先在甲苯中進行共沸除水處理,無需干燥。隨后,加入硼酸(115g)和濃硫酸(415g),在105-110℃下加熱反應4-8小時。反應完成后,將反應混合物緩慢倒入冷水中,并用乙酸乙酯進行萃取。最后,通過減壓蒸餾除去溶劑,得到目標產物8-氯-11-(1-甲基哌啶-4-亞甲基)-6,11-二氫-5H-苯并[5,6]環(huán)庚[1,2-b]吡啶,為固體形式,產率為85%,純度為99.5%(通過OAB和HPLC測定)。
