向苯甲酰乙酸乙酯(2g,10.41mmol)的乙醇(15mL)溶液中加入乙醇鈉(7mL,18.7mmol)和化合物(CAS:17356-08-0)(1.18g,15.61mmol)。將反應(yīng)混合物在回流條件下攪拌過夜。反應(yīng)完成后,減壓蒸餾除去溶劑。將殘余物溶于水,用酸調(diào)節(jié)pH至3,然后用乙酸乙酯(3×20mL)萃取。合并有機(jī)相,用鹽水洗滌,無水硫酸鈉干燥。粗產(chǎn)物通過快速柱色譜法純化,以二氯甲烷/甲醇(2.5%)為洗脫劑,得到2-巰基-6-苯基-4-羥基嘧啶(500mg,2.44mmol),為白色固體,收率為24%。
