以4-氟苯酚和三氯甲烷為原料合成5-氟水楊醛的一般步驟如下: 步驟A: 向含有NaOH(78g,1950mmol)的水(400mL)溶液中加入4-氟苯酚(50g,446.43mmol)的水(200mL)和乙醇(150mL)混合溶液。將混合物加熱至70℃后,通過加料漏斗緩慢滴加三氯甲烷(110mL)(約2小時(shí)完成滴加),并在該溫度下持續(xù)攪拌反應(yīng)混合物過夜(約16小時(shí))。反應(yīng)完成后,將混合物冷卻至室溫,用3M HCl溶液酸化。隨后,將反應(yīng)混合物在飽和食鹽水和二氯甲烷之間進(jìn)行分配,分離有機(jī)層,用無水硫酸鈉干燥,過濾并濃縮。粗產(chǎn)物通過硅膠柱色譜法純化(洗脫劑:3%乙酸乙酯/己烷),得到5-氟水楊醛13.6g(收率22%,白色固體)。
